Only judge the artist/band based on the music that was released in that decade, e.g., if you select the Cure for the 90s, it would only be based on Wish, Wild Mood Swings, and some of the other random things they put out during that period.
1950s: Buddy Holly
1960s: The Beatles
1970s: Pink Floyd
1980s: The Smiths
1990s: R.E.M. (I think Out of Time, Automatic for the People, and Up, are excellent albums)
2000s: Death Cab for Cutie
1950s: Buddy Holly

Now this is a great thread! Nice idea, No I in Don! Anyway, here I go...
50s: Nat King Cole
Runner up: Frank Sinatra
60s: The Beatles
Runner up: Patsy Cline
70s: Ramones
Runner up: Pink Floyd/The Who/Queen/Patti Smith
80s: The Smiths/The Clash
Runner up: The Chameleons/The Cure/Talking Heads/Kate Bush
90s: Radiohead
Runner up: R.E.M
00s: Interpol
Runner up: Arcade Fire/Of Montreal
I know I cheated, sue me. But it's hard narrowing this thing down when there's so many bands and musicians that I absolutely love.

50's - Frank Sinatra: the comeback, the Capitol years, the Ocsar, the beginnings of the Vegas years
60's - The obvious choice is the Beatles, but I feel honorable mention should be the Velvet Underground for being one of the first true alternative bands.
70's - David Bowie: Ziggy Stardust, Station to Station, Low, the Thin White Duke.
80's - The Clash: the only band that mattered. London Calling and Combat Rock embraced media and new technologies with an eye to the future. Green Day would not be on Broadway without them.
90's - Radiohead: un-grunge experimentalists saved those of us who did not like Nirvana. Three great albums in the nineties.
2000's - The Shins: because they could change your life according to Zach Braff. Runner up would be Interpol. Oh, and then there's Spoon...

50's - I honestly can't form an opinion on this, I know only Elvis and Buddy Holly.
60's - The Beatles. If you have any other answer, you're just wrong.
70's - Led Zepplin
80's - The Smiths
90's - Radiohead
00's - The Shins
That was ridiculously hard. I had to just go with first instinct, if I thought about it for too long I would have about 10 artists for each decade (except the 50's. I fail at 50's knowlege.)

My toughest decade was the 90s. There were good bands that came out in the 90s, but I guess I didn't feel a personal connection to most of them. All of my favorites from the 80s put out music (e.g., Morrissey, The Cure, Depeche Mode), but they weren't very prolific. I almost went with Depeche Mode because they technically released three excellent albums in the 90s (Violator, Songs of Faith & Devotion, & Ultra), but I felt like Violator was cheating because the album was recorded in 1989 and the first single "Personal Jesus" hit radio before the new year.

Zach Braff... the last time he was even culturally relevant was 2004.
50's: The California Gold Rush Singers
60's: "Honest" Abe Lincoln & The Emancipators
70's: The Ringers (Featuring Alexander Graham Bell)
80's: Jack & The Rippers (First-Wave British Invasion)
90's: The Roughriders
00's: Sousa
